INDICATION OF DOMPERIDONE
This medicine is useful to treat & relieve nausea & vomiting due to any cause, mention below
- Nausea or vomiting due to any medicine
- After operation
- Due to radiation
- Uraemia
- hepatitis
- peptic ulcer
- reflux oesophagitis
- diabetic gastroparesis
- functional dyspepsia
- adjuvant in vertigo with nausea
CONTRAINDICATION
It is more important than indication. We must know the conditions in which we should avoid this medicine.
- Gastro intestinal obstruction
- Gastro intestinal perforation
- Hypersensitivity to this molecule
- Extrapyramidal symptoms
- Phaeochromocytoma
- Epilepsy
DOSAGE OF DOMPERIDONE
Dosage may depend on severity of complain, age & weight. Kindly follow your doctor advise for dosage.
Adult
10-20mg three to four time in a day before meal
Childen
0.3mg/kg body weight per dose
10kg child – 3mg three time in a day
20 kg child – 6mg three time in a day
Onset of effect of domperidone starts within 1 hour after taking it & effect last for approx 6 hours after taking it.
SIDE-EFFECT
Each & every molecule which have effect also have side-effect. But, it not occurs to everyone. It occurs to only who have sensitivity to particular molecule.
If adverse effect occurs, that may be one of below
- Drowsiness & extra pyramidal reaction
- Convulsion
- Galactorrhea (raised serum prolactin)
- Muscle spasm
- Rashes over body
- Breast enlargement
- Constipation or diarrhoea
- Hypertensive crisis in patient with pheochromocytoma
SPECIAL PRECAUTION
- Use cautiously in kidney disease
- Cautiously use in predisposition to cardicac arrythmias or hypocalaemia
MECHANISM OF ACTION
Domperidone antagonises D2 receptor & it antagonizes the inhibitory effect of dopamine. It results into stimulation of gastric muscle contraction. Due to that we get relief in nausea & vomiting.
